AKY-1189 has demonstrated preclinical superiority versus Padcev in low-Nectin-4 models and early human imaging across multiple cancers, while AKY-2519 delivered 92% tumor-growth inhibition in NSCLC xenografts and supports a non-PSMA entry point into mCRPC and lung cancer. Aktis Oncology is viewed negatively because its valuation depends heavily on unproven clinical-stage assets, and the biggest near-term risk is poorer-than-expected results or failure of AKY-1189 and AKY-2519 to show differentiated activity and tolerability. Although dosimetry for [177Lu]Lu-AKY-1189 showed low median absorbed dose to the kidney at 0.30 Gy/GBq and to bone marrow at 0.01 Gy/GBq, the company still faces substantial execution risk from clinical delays, competitor products, isotope supply or manufacturing constraints, and reimbursement uncertainty. Its DCF framework also embeds only 20% probability of approval for each lead program and highlights long-term dilution risk, underscoring limited fundamental cushion if the 1Q27 UC readout or later studies disappoint.
